Serum antibody response to Bacteroides fragilis in experimental sinusitis

Summary

Maxillary sinusitis was induced in New Zealand white rabbits by local inoculation of 106 colony-forming units ofBacteroides fragilis NCTC 9343, and the serum IgG, IgA and IgM antibody responses to cell wall antigens were studied. Prior to inoculation, and one, two, three and four weeks after induction, serum samples were obtained and analysed for antibodies to the lipopolysaccharide and the capsular polysaccharide. Capsular polysaccharide fromB. fragilis ATCC 23745 was used as control. The rise in IgG activity against NCTC 9343 capsular polysaccharide and lipopolysaccharide was most marked and sustained throughout the four weeks. The increases in IgA concentration were moderate and sometimes transient, and a more pronounced IgA increase was seen with IFA than with EIA. The IgM peak levels were weak and usually declined within two to three weeks. The development of antibody to the lipopolysaccharide was similar to that of the NCTC 9343 capsular polysaccharide antibodies, though somewhat delayed in time. No significant increase in antibody to the control capsular polysaccharide was seen.

Zusammenfassung

Bei weißen Neuseeland-Kaninchen wurde durch lokale Inokulation von 106 koloniebildenden Einheiten vonBacteroides fragilis NCTC 9343 eine Sinusitis maxillaris induziert; die Reaktion der Serum IgG-, IgA- und IgM-Antikörper gegen Zellwandantigene des Erregers wurde untersucht. Serumproben wurden vor der Inokulation und eine, zwei, drei und vier Wochen nach Setzen der Sinusitis entnommen und auf Antikörper gegen das Lipopolysaccharid und Kapselpolysaccharid des Teststammes geprüft. Als Kontrolle diente Kapselpolysaccharid vonB. fragilis ATCC 23745. Während der vier Untersuchungswochen kam es zu einer ausgeprägten und anhaltenden IgG-Reaktion gegen das Kapselpolysaccharid und Lipopolysaccharid des Stammes NCTC 9343. Die IgA-Konzentrationen stiegen mäßig an; bei einigen Tieren war der Anstieg vorübergehend; die IgA-Reaktion war mit IFA ausgeprägter als mit EIA. IgM zeigte nur gering ausgeprägte Reaktivität mit niedrigen Spitzenkonzentrationen, die bereits nach zwei bis drei Wochen wieder abfielen. Mit gewisser zeitlicher Verzögerung stellte sich eine ähnliche Antikörperreaktion auf das Lipopolysaccharid vonB. fragilis NCTC 9343 ein. Gegen das Kapselpolysaccharid des Kontrollstammes war keine ausgeprägte Antikörperreaktion festzustellen.
